If you read deadly fortune (the dmc 4 novels, i found it online but the translation was a little rough. Still got the point across) it gives a better insight into Dante's feelings toward Nero throughout the game.

IIRC, Dante knew Nero was related to him even before it was confirmed through the Yamato. He could sense it, like his spidey senses were tingling or something. That sense reminded him a lot of Vergil, and he kinda assumed from there - obviously Nero isnt his kid, so he has to be somebody's kid, and that somebody was very clearly somebody like Dante himself. Somebody like Vergil, perhaps? Although it was still just a theory since he meets Nero after he's gotten Vergil killed (twice).

Either way, he did know Nero was related to him in some way or another.

Thats going off memory tho i havent read deadly fortune in a while.

I bet the second Dante sees Nero, his attitude and also what Nero really stands for in DMC4, he IMMEDIATELY puts 2 and 2 together, figures out whose son Nero is, and, like, the entire reason Dante trolls Nero and fucks around so much but also sneakingly mentors him and trusts him to figure the Order out on his own in a way only uncle can is because Dante just knows, even before Yamato pretty much confirms it for him. But he also can't help himself trolling Nero and getting into fights because he just sees Vergil parts in him
